WORK DONE IN BRIEF Ph.D. thesis on the topic entitled “STUDIES ON LUCIFERASE ENZYMES FOR THEIR APPLICATION IN ASSESSING HYGIENE STATE OF SELECTED FOOD SAMPLES”

Luciferase enzyme from indigenously available fireflies (Asymmetricata humeralis walker) was extracted which has been not characterized till date. Purification of firefly luciferase enzyme from these luminescent insect was carried out which was utilized for hygiene monitoring, rapid determination milk and fish freshness. Further, an In-house avalanche photodiode and photomultiplier tube based bioluminometer was fabricated for the development of optical biosensor.

WORK DONE IN BRIEF FOR Ph.D. thesis on the topic entitled “STUDIES ON LUCIFERASE ENZYMES FOR THEIR APPLICATION IN ASSESSING HYGIENE STATE OF SELECTED FOOD SAMPLES”

Rapid prescreening of hazardous materials (Heavy metals and pesticides) present in food and water samples was carried out using whole cell bioluminescent bacteria having in vivo luciferase activity (P. leiognathi) after immobilization. For this, potent bioluminescent strain was isolated from marine mussels and were characterized biochemically and through 16S rRNA sequencing. Parametric optimization of growth and nutrient conditions were carried out for enhanced luminescence emission from P. leiognathi. Further, luciferase enzyme from P. leiognathi was extracted and biochemical parameters were optimized
